The present study investigates the effect of insulin treatment on AT1 receptor expression and ANG II-induced stimulation of Na/H exchanger (NHE) and Na-K-ATPase (NKA) in opossum kidney (OK) cells, a proximal tubule cell line.
The whole cell lysate and membrane showed similar insulin-induced increase in the AT1 receptor protein expression, which was blocked by genistein (100 nM), a tyrosine kinase inhibitor, and cycloheximide (1.5 microg/ml), a protein synthesis inhibitor.

These effects of dexamethasone were blocked by cycloheximide, a protein synthesis inhibitor.

Calnexin binding persists for several hours after translation is stopped with cycloheximide, suggesting that the beta (2) subunit undergoes repeated post-translational calnexin-assisted folding attempts.

The B subunits of V-ATPases from mammals and tobacco hornworm have been shown to bind actin filaments.
Screening null yeast or null yeast transformed with wild-type or mutant B subunits with sub-lethal doses of various drugs revealed that yeast containing the mutant B subunits were more sensitive to cycloheximide and wortmannin than those transformed with wild-type B subunits.

Hence, co-sorting of the H,K-ATPase beta subunit with the Na,K-ATPase alpha (1) subunit to the basolateral membrane in MDCK cells may determine the differential distribution of the beta subunit in these two cell types.

Hepatology. 2009 Dec;50(6):1871-83.

This restoration was not observed when cells were also treated with cycloheximide, thus confirming that proteasomal degradation in this setting was restricted to newly synthesized Pontin.
Reptin and Pontin are related ATPases associated with stoichiometric amounts in several complexes involved in chromatin remodeling, transcriptional regulation, and telomerase activity.

Cycloheximide did not affect resensitization, but bafilomycin A (1), an inhibitor of vacuolar H (+)-ATPases, abolished resensitization.
