The intrathecal effect of 0.1 to 10 microg of RP-67,580 (3aR,7aR)-7,7-diphenyl-2 [1-imino-2 (2-methoxyphenyl)-ethyl]++ +perhydroisoindol-4-one hydrochloride, CP-96,345 (2S,3S)-cis-(2 (diphenylmethyl)-N-[(2-methoxyphenyl) methyl]-1-azabicyclo [2.2.2] octan-3-amine), SR-140,333 (S)-(1- inverted question mark2-[3-(3,4-dichlorophenyl)- 1-(3-isopropoxyphenylacetyl) piperidin-3-yl] ethyl inverted question mark-4-phenyl-1 -azonia-bicyclo [2.2.2.]-octane,chloride), all neurokinin (NK) 1-receptor antagonists, SR-48,968 (S)-N-methyl-N [4-(4-acetylamino-4-[phenylpiperidino)-2-(3,4-dichlorophen yl)-butyl] benzamide, a tachykinin NK2 receptor antagonist and SR-142,801 (S)-(N)-(1-(3-(1-benzoyl-3-(3,4-dichlorophenyl) piperidin-3-yl) propyl)-4-phenylpiperidin-4-yl)-N-methyl acetamide, a tachykinin NK3 receptor antagonist, and of their respective inactive enantiomers on thresholds of vocalization due to a mechanical stimulus in mononeuropathic (sciatic nerve ligature) and diabetic rats, was examined. |

9718274 | Matuszek MA, Zeng XP, Strigas J, Burcher E: An investigation of tachykinin NK2 receptor subtypes in the rat. Eur J Pharmacol. 1998 Jul 3;352(1):103-9. Neurokinin A, neuropeptide gamma, [Lys5,MeLeu9,Nle10] NK (4-10), SR 48968 [(S)-N-methyl-N [4-(4-acetylamino-4-phenylpiperidino)-2-(3,4-dichlorophen yl) butyl] benzamine], GR 94800 [PhCO- - -DTrp- -DPro-Pro-Nle-NH2] and MEN 10627 [cyclo (Met-Asp-Trp- -Dap- cyclo (2beta-5beta)] displayed high affinity (pIC50 8.4-9.5) as competitors, with no significant difference in potency between these four tissues. [Lys5,MeLeu9,Nle10] neurokinin A-(4-10) contracted the isolated fundus (EC50 117 nM) and bladder (EC50 10 nM) and these responses were similarly inhibited by the tachykinin NK2 receptor antagonists, SR 48968 and MEN 10627 (pA2 values 7.6-8.2). |

9851584 | Hsu TH, Lai YL, Kou YR: receptor antagonists attenuate wood smoke-induced bronchoconstriction in guinea pigs. Eur J Pharmacol. 1998 Nov 6;360(2-3):175-83. The smoke-induced changes in RL and Cdyn were significantly attenuated by pretreatment with atropine, CP-96,345 [(2S,3S)-cis-2-(diphenylmethyl)-N-((2-methoxyphenyl)-methyl)-1-aza bicyclo (2.2.2.)-octan-3-amine; a tachykinin NK1 receptor antagonist], and SR-48,968 [(S)-N-methyl-N (4-(4-acetylamino-4-phenylpiperidino)-2-(3,4-dichlorophen yl)-butyl) benzamide; a tachykinin NK2 receptor antagonist] in combination, atropine alone, and SR-48,968 alone, but were not significantly affected by pretreatment with the inactive enantiomers of CP-96,345 and SR-48,968, CP-96,345 alone, indomethacin (a cyclooxygenase inhibitor), and MK-571 [((3-(3-(2-(7-chloro-2-quinolinyl) ethenyl) phenyl ((3-dimethyl amino-3-oxo-propyl) thio) methyl) propanoic acid; a receptor antagonist]. |

Eur J Pharmacol. 1996 Mar 7;298(2):113-20. Similarly, the bradykinin B2 receptor antagonist Hoe 140 (D-Arg-[Hyp3,Thi5,DTic7,Oic8]-bradykinin) (0.6 mg/kg), the PAF receptor antagonist WEB 2086 (3-(4-(2-chlorophenyl)-9-methyl-6H-thieno-(3,2f) (1,2,4)-triazolo-(4,3-a) (1,4)-diazepine-2-yl)-(4-morpholinyl)- (20 mg/kg), the tachykinin NK1 receptor antagonist SR 140333 ((S) 1-(2-[3-(3,4-dichlorophenyl)-1-(3-iso-propoxyphenyl acetyl) piperidin-3-yl] ethyl)-4-phenyl-1-azoniabicyclo [2.2.2] (0.5 mg/kg), the tachykinin NK2 receptor antagonist SR 48968 ((S)-N-methyl-N [4-(4-acetylamino-4-phenylpiperidino)-2-(3,4-dichlorophen yl) butyl] benzamide) (0.5 mg/kg) and the synthase inhibitor N omega-nitro- methyl ester (10 mg/kg) had no significant effect on the mean arterial blood pressure changes induced by Phoneutria nigriventer venom. |
