In the present study, we investigated the modulatory effects of eugenol on NF-kappaB signaling in a rat model of gastric carcinogenesis induced by N-methyl-N (')-nitro-N-nitrosoguanidine (MNNG) by analysing the expression of nuclear factor-kappaB (NF-kappaB) family members ((NF-kappaB (p50 and p65), inhibitor of kappaB alpha (IkappaBalpha), phosphorylated IkappaBalpha (p-IkappaBalpha), IkappaB kinase beta (IKKbeta)) and the NF-kappaB target genes that promote (e.g., cyclin D1, cyclin B and PCNA) or inhibit (e.g., p53, p21, and Gadd45) cell proliferation and cell survival.

Isoeugenol inhibited NF-kappaB-dependent transcriptional activity and DNA-binding activity by decreasing the nuclear translocation of p65, which is a component of NF-kappaB.
The isoeugenol analogues eugenol and allylbenzene also inhibited LPS-induced NF-kappaB signaling and iNOS expression, albeit with less potency than isoeugenol.
