Carvacrol causes concentration-dependent increases in the intracellular [Ca (2+)] of native cerebral artery endothelial cells and is more potent (EC (50) = 34 microM) than the TRPA1 agonist allyl isothiocyanate (EC (50) = 400 microM) or the TRPV3 agonist eugenol (EC (50) = 2.3 mM).

In the present study we demonstrate by patch clamp whole cell recordings from Schneider 2 and Drosophila photoreceptor cells that carvacrol, a known activator of the thermoTRPs, TRPV3 and TRPA1 is an inhibitor of the Drosophila TRPL channels, which belongs to the TRPC subfamily.
We also show that additional activators of TRPV3, thymol, eugenol, cinnamaldehyde and menthol are all inhibitors of the TRPL channel.

Carvacrol, eugenol and thymol are major components of plants such as oregano, savory, clove and thyme.
In addition, carvacrol activates and rapidly desensitizes TRPA1, which may explain the pungency of oregano.
