19349754 Kim JH, Lee CH: Rengyolone inhibits apoptosis via etoposide-induced caspase downregulation. J Microbiol Biotechnol. 2009 Mar;19(3):286-90.

The regulating compounds were purified from the ethyl acetate extract by silica gel column chromatography and HPLC.
This compound showed inhibitory activity on caspase-3 induction, a major protease of the apoptosis cascade, with an IC50 value of 38.96 microM after 8 h of etoposide treatment in U937 cells.

From a silica gel column chromatograpy of this extract, we obtained four fractions (F-2, F-4, F-5 and F-7) having apoptosis-inducing activity.
These fractions induced DNA ladder and caspase-3 activation during apoptosis in colon 26 tumor cells.

By using ethyl acetate extraction and silica gel chromatography, an active fraction of THL named as EAS5 was isolated.
The THL-induced apoptosis was accompanied with caspase-3 activation and decrease of PML-RARalpha fusion protein.

The methanolic extract was sub-fractionated on a silica gel G column using a combination of non-polar and polar solvents and eleven fractions were obtained.
Finally, the mechanism of cell death was investigated by measuring the levels of Bcl2, caspase 3 and DNA fragmentation.

Oxypeucedanin-induced cell death was associated with significant increases in apoptosis and cleaved caspase-3 and poly-(ADP-ribose) polymerase.
MATERIAL AND METHODS: Oxypeucedanin (C (16) H (14) O (5), mw: 286) was isolated through silica gel chromatography and characterized by NMR.
