We gave nicotine to adolescent rats (postnatal days, PD, 30-47), simulating plasma levels in smokers, and then examined the subsequent effects of nicotine given again in young adulthood (PD 90-107), focusing on 5-HT (1A) and 5-HT (2) receptors and the 5-HT transporter during nicotine treatment (PD 105) and withdrawal (PD 110, 120, 130), and long-term changes (PD 180).

These changes in heart rate variability were associated with increased 5-HT (1A) receptor binding in the raphe obscurus (P = 0.04) and increased nicotinic receptor binding in the raphe obscurus and vagal complex (P < 0.05) in the nicotine-exposed animals compared with controls (n = 6).

Additionally, 8-OH-DPAT- and clonidine-induced hypothermia was tested in vehicle- and nicotine-pretreated mice, as a measure of 5-HT (1A) and alpha (2)-adrenoceptor function, respectively.

Nicotine was infused in pregnant rats via implanted osmotic minipumps (6.5mg/kg per day) from gestational day 3 (G3) until G21, the day of delivery.
