The in vitro interaction of 3,4,5,6-tetrachloro-1,2-benzoquinone (o-chloranil) with rat liver glutathione S-transferases (GST) was studied, using reduced glutathione (GSH) and 1-chloro-2,4-dinitrobenzene (CDNB) as substrates. o-Chloranil inhibited the GST activity in crude extracts in a dose dependent manner, while 3,6-dichloro-2,5-dihydroxy-1,4-benzoquinone did not.

The interaction of quinones, o-chloranil and chlorophenoxyalkyl acids with the GST activity, in extracts from three different macro-invertebrates, revealed an inhibition which was quite similar to that previously found for rat liver GST.

J Biol Chem. 1994 Oct 28;269(43):26890-7.

The mode of inactivation of glutathione S-transferase isoenzyme 3-3 from rat by the active site-directed inhibitor 2-(S-glutathionyl)-3,5,6-trichloro-1,4-benzoquinone (GSTCBQ) has been investigated by a combination of site-specific mutagenesis and mass spectrometric analysis of the sites of reaction of the reagent with the enzyme.
