18339394 | Cao X, Hu G, Huo L, Zhu X, Li T, Powell J, Ito Y: Stationary phase retention and preliminary application of a spiral disk assembly designed for high-speed counter-current chromatography. J Chromatogr A. 2008 Apr 25;1188(2):164-70. Epub 2008 Mar 4. The results indicated that the spiral disk assembly can produce excellent retention of stationary phase for moderately polar organic-aqueous solvent systems, such as chloroform- -water (4:3:2) and hexane-ethyl - -water (1:1:1:1) by pumping lower mobile phase from head (H) to tail (T), and upper mobile phase from tail (T) to head (H) even at a high flow-rate (8 mL/min, Sf> 70%), regardless of whether the inlet is at the inner or outer terminal of the channel. Acceptable resolutions were achieved when it was used for the separation of peptides such as -Tyr and Val-Tyr, and proteins including cytochrome c and myoglobin, lysozyme and myoglobin, and fresh chicken egg-white proteins. |

That is, the precipitated activated sludge was washed with TENP and PBS buffer, followed by using lysozyme and TRIzol to direct lysis of microbial cells, chloroform to remove protein and most of the DNA from bacterial lysate, to precipitate nucleic acid and DNase I to hydrolyze residual DNA. |

Epub 2009 Jan 17. Nanoparticles containing the model protein lysozyme have been produced using microemulsion and nanoprecipitation methods. Freeze-drying water in oil emulsions, with chloroform as the organic solvent, followed by washing of excess surfactant led to the production of nanoparticles having approximately 300 nm mean size. |

For disintegration and extraction, we combined primarily mild techniques: enzymatically we used combinations of lysozyme and lipase for bacteria as well as lyticase and lipase for yeasts. For extraction we used a -chloroform mixture stabilized efficiently with butylated hydroxytoluene and |

The effects of extraction buffers and lysis treatments (lysozyme, (SDS), sonication, mechanical mill and thermal shock) on yield and purity of the total DNA extracted from activated sludge were investigated. Cetyltrimethyl ammonium and /chloroform were used to remove proteins and polysaccharides from crude DNA. |
