Our assay is based on the differential partitioning of substrate and product following a single chloroform/methanol extraction.
The substrate may be delivered using either fatty acid-free albumin or detergent/lipid mixed micelles, and we have found that the use of albumin rather than detergent micelles allows one to detect lipid interactions with the enzyme that might otherwise go unnoticed.

Using chloroform/methanol separation of rat small intestine homogenates into lipid fractions and aqueous and sedimented protein fractions and measuring cell death caused by those fractions, we found that the cytotoxic factors are lipid in nature.
Addition of albumin, a lipid binding protein, prevented cell death, unless the albumin was previously digested with protease.

In addition to bilirubin, Cuc B, D, and E increased the binding constant values for warfarin to albumin, whereas they did not affect the binding of other ligands of site I such as chloroform and salicylate.
Their binding to albumin may control their diffusion and consequently their biological effects.
