Investigations to determine the inhibitory activity on the Ca (2+)-transport-ATPase of human erythrocyte membranes were performed with various compounds of toxicological significance, mostly chlorinated and mainly used as biocides, such as phenol, 4-chlorophenol, 2,4-dichlorophenol, 2,6-dichlorophenol, 3,4-dichlorophenol, 2,3,4-trichlorophenol, 2,3,4,5-tetrachlorophenol, pentachlorophenol (PCP), captan, folpet, captafol, (+)-camphene, toxaphene, dichlorodiphenyltrichloroethane (DDT), lindane, endrin, dieldrin, alpha-endosulfan, beta-endosulfan, paraquat, diallate, 2,4-dichlorophenoxyacetic acid (2,4-D) and 2,4,5-trichlorophenoxyacetic acid (2,4,5-T).

Endosulfan stereoisomer I (E-I) inhibited the activity of calmodulin-dependent Ca2+-ATPase to an extent of 15-55% in a concentration range of 1-20 microM under in vitro conditions without significantly affecting the basal enzyme activity.

The marked sensitivity of mitochondrial Mg2+ ATPase to endosulfan is suggestive of the potential for endosulfan to interfere markedly with various energy requiring processes in the fish body.

Endosulfan might enhance calcium concentration in the spermatogenic cells and restrained Na+ K+ -ATPase and Ca2+ Mg2+ -ATPase activities that showed endosulfan might induce spermatogenic cells in the testicle to occur apoptosis (p < 0.01), to cause the function of transporting Ca2+ to be limited, reduce the ability of excluding calcium and send Ca2+ concentration to hoist sustainingly,i.e. calcium overload in the cells.

Results indicated significant reduction in the growth performance, survival, and activities of lactate dehydrogenase (liver and muscle), malate dehydrogenase (liver and muscle), enzymes of protein metabolism (aspartate amino transferase in liver and alanine amino transferase in liver and muscle), acetyl choline esterase (brain), alkaline phosphatase activity (liver), and ATPase (gill) enzymes of group fed control diet and exposed to endosulfan.

The partially purified Ha-Pgp ATPase was reconstituted into proteoliposomes and it was found that some insecticides, namely, monocrotophos, endosulfan, cypermethrin, fenvalerate, and methylparathion, stimulated the ATPase activity.

We studied the effects of four commonly used insecticides (methylparathion, endosulfan, cypermethrin and fenvalerate) on P-glycoprotein isolated from multidrug-resistant cells.
All the pesticides stimulated P-glycoprotein ATPase activity, with maximum stimulation of up to 213% in a detergent-solubilized preparation, and up to 227% in reconstituted liposomes.
