19351244 | Yin Y, Liu X, Li B, Ma Z: Characterization of sterol demethylation inhibitor-resistant isolates of Fusarium asiaticum and F. graminearum collected from wheat in China. Phytopathology. 2009 May;99(5):487-97. In this study, sensitivities of 159 F. asiaticum and F. graminearum isolates to a benzimidazole fungicide carbendazim (MBC) and to sterol demethylation inhibitors (DMIs) tebuconazole and prochloraz were determined. Analysis of two 14alpha-demethylase (cyp51) homologous genes (cyp51A and cyp51B) showed that the F. asiaticum isolates could be distinguished from F. graminearum isolates based on the sequence of cyp51A. |

Azole antifungals are central to therapy and act by inhibiting a cytochrome P450, sterol 14-demethylase and blocking normal sterol synthesis. Several showed equivalent or greater inhibitory effects to those against Candida albicans, and bactericidal activity was demonstrated for four compounds, econazole, miconazole and tebuconazole. |

18257249 | Xiao M, Yang JY, Xiao WJ, Yang S: [Application of binding spectra in DMIs fungicide screening] . Sheng Wu Gong Cheng Xue Bao. 2007 Nov;23(6):1129-34. In order to establish a fast and accurate method for novel DMIs fungicide screening, 14alpha-demethylase of Magnaporthe grisea expressed in E. coli was used as target enzyme and the DMI fungicides diniconazole, tebuconazole, triadimenol and triadimefon were used as representative fungicides, the effects of enzyme activity, enzyme purity and concentration on the binding spectra were investigated. |

Pest Manag Sci. 2003 Dec;59(12):1333-43. The isolates tested represent the wide baseline sensitivity to the azole fungicide tebuconazole described previously. One isolate with intermediate sensitivity to azoles over-expressed CYP51, encoding cytochrome P450 sterol 14alpha-demethylase from M graminicola. |

An Ustilago maydis biosynthesis mutant (A14) which is partially blocked in sterol 14alpha-demethylase (P45014DM) activity is described. The mutant was also found to be hypersensitive to the azole fungicides penconazole and tebuconazole. |
