An i.p. administration of rats with piperine (100 mg/kg) and piperonyl butoxide (400 mg/kg) produced a significant decrease in hepatic cytochrome P-450, and activities of benzphetamine N-demethylase, aminopyrine N-demethylase and aniline hydroxylase 1 hr after the treatment.

6814016 Fisher CW, Mayer RT: Characterization of house fly microsomal mixed function oxidases: inhibition by juvenile hormone i and piperonyl butoxide. Toxicology. 1982;24(1):15-31.

The inhibition of these reactions by juvenile hormone I (E, E cis methyl 10,11-epoxy-7-ethyl-3, 11-dimethyl-2, 6-tridecadienoate, JH-I) was competitive for the N-demethylase, epoxidase and the formation of the PB metabolite-complex.

However, both pargyline and deprenyl also inhibited cytochrome P-450 mediated aminopyrine N-demethylase activity in brain slices.
Other cytochrome P-450 inhibitors, namely, piperonyl butoxide and SKF 525A were found to offer protection against MPTP induced neurotoxicity in slices without affecting monoamine oxidase activity.

To investigate the selectivity and safening action of the sulfonylurea herbicide pyrazosulfuron-ethyl (PSE), pyrazosulfuron-ethyl O-demethylase (PSEOD) activity involving oxidative metabolism by cytochrome P-450 was studied in rice (Oryza sativa L cv Nipponbare) and Cyperus serotinus Rottb.

A significant decrease in aminopyrine N-demethylase was also noticed after pulegone administration.
Pretreatment of rats with phenobarbital (PB) or diethylmaleate (DEM) potentiated the hepatotoxicity caused by pulegone, whereas, pretreatment with 3-methylcholanthrene (3-MC) or piperonyl butoxide protected from it.

The synergists piperonyl butoxide and S,S,S-tributylphosphorotrithioate increased t-permethrin toxicity four- and threefold (at the LC50) in the UF and ARS colonies, respectively.

The toxicity of spinosad could be synergised by piperonyl butoxide (PBO) and triphenylphosphate (TPP), but not by diethyl maleate (DEM).

Binding to hepatic microsomal protein was decreased by pretreatment with phenobarbital or piperonyl butoxide, but not significantly with allopurinol. 6-Thiopurine at therapeutic doses had no apparent negative effects on either hepatocellular glutathione levels or hepatic microsomal aminopyrine N-demethylase activity during this time period.

In vitro, the concentration of methoxsalen inhibiting by 50% monooxygenase activities ranged from 10 microM (for benzo (a)-pyrene and hexobarbital hydroxylases] to 25 microM for 7-ethoxy-coumarin deethylase and aminopyrine demethylase); these values were in the range of those observed with SKF 525-A (1-50 microM) or piperonyl butoxide (10-100 microM) but much lower than those for cimetidine (100-500 microM).
