Folate targeted gene delivery vectors showed enhanced accumulation in folate receptor expressing tumor model.
Folate mediated targeting was induced by conjugating poly (ethylene glycol)-folate (PEG-FA) on histidine modified chitosan polymer having low molecular weight of 15 kDa and high degree of quaternisation (HTFP15-H).

Folate-conjugated unimolecular micelles based on amphiphilic hyperbranched block copolymer, Boltorn H40-poly (l-aspartate-doxorubicin)-b-poly (ethylene glycol)/FA-conjugated poly (ethylene glycol) (H40-P (LA-DOX)-b-PEG-OH/FA), were synthesized as a carrier for tumor-targeted drug delivery.
Cellular uptake of the H40-P (LA-DOX)-b-PEG-OH/FA micelles was found to be higher than that of the H40-P (LA-DOX)-b-PEG-OH micelles because of the folate-receptor-mediated endocytosis, thereby providing higher cytotoxicity against the 4T1 mouse mammary carcinoma cell line.

Gold (Au) nanoparticles (NPs) stabilized with a monolayer of folate-conjugated poly (L-aspartate-doxorubicin)-b-poly (ethylene glycol) copolymer (Au-P (LA-DOX)-b-PEG-OH/FA) was synthesized as a tumor-targeted drug delivery carrier.
Cellular uptake of the Au-P (LA-DOX)-b-PEG-OH/FA micelles facilitated by the folate-receptor-mediated endocytosis process was higher than that of the micelles without folate.

The folate-conjugated micelles can be internalized by the cancer cells via folate-receptor-mediated endocytosis; thus, they exhibited enhanced cell uptake and cytotoxicity.
The inner hydrophobic layer was composed of random copolymer of poly (epsilon-caprolactone) and poly (malic acid) (PMA-co-PCL) segments, while the outer hydrophilic shell was composed of poly (ethylene glycol) (PEG) segments.

Biomaterials. 2010 Mar 25.

To develop a drug delivery system with enhanced efficacy and minimized adverse effects, we synthesized a novel polymeric nanoparticles, (YCC-DOX) composed of poly (ethylene oxide)-trimellitic anhydride chloride-folate (PEO-TMA-FA), doxorubicin (DOX) and superparamagnetic iron oxide (Fe (3) O (4)) and folate.
YCC-DOX showed the anticancer efficacy and specifically targeted folate receptor (FR)-expressing tumors, thereby increasing the bioavailability and efficacy of DOX.

The study developed cytotoxic macromolecular conjugates that specifically target the folate receptor and deliver the drug into cell cytoplasm.
The anticancer agent paclitaxel was conjugated to human serum albumin (HSA) and this drug-albumin conjugate was further equipped with folic acid, linked via an extended poly (ethylene glycol) spacer.
