N (6)-Isopentenyladenine was the predominant cytokinin in the medium of all five strains, and its increased production was correlated with the degree of pathogenicity; however, the virulent strains also produced 8-13 times more cis-zeatin [6-(4-hydroxy-3-methyl-cis-2-butenylamino) purine] than the avirulent strain.

There was a much greater difference in activity (> 50-fold) between trans- and cis-zeatin than between trans-isozeatin [6-(4-hydroxy-2-methyl-trans-2-butenylamino) purine] and cis-isozeatin [6-(4-hydroxy-2-methyl-cis-2-butenylamino) purine], the latter being less active than cis-zeatin and trans-isozeatin.

One of these has been identified as 6-(4-hydroxy-3-methyl-cis-2-butenylamino)-2-methylthiopurine (2-methylthio-cis-zeatin, c-ms (2) io (6) Ade). The elution volumes of the other two fractions correspond to those of authentic 6-(4-hydroxy-3-methyl-2-butenylamino)-9-beta-d-ribofuranosyl-purine and 6-(3-methyl-2-butenylamino)-9-beta-d-ribofuranosylpurine, indicating the presence of trace amounts of these two ribonucleosides.

One constituent is now identified as cis-zeatin [6-(4-hydroxy-3-methyl-cis-2-butenylamino) purine].

16659634 Einset JW, Swaminathan S, Skoog F: Ribosyl-cis-zeatin in a Leucyl Transfer RNA Species from Peas. Plant Physiol. 1976 Aug;58(2):140-142.

The cytokinin has been identified as ribosyl-cis-zeatin, 6-(4-hydroxy-3-methyl-cis-2-butenylamino) -9-beta-d-ribofuranosylpurine.

Besides the expected 6-[(3-methylbut-2-enyl) amino]-9-(beta-D-ribofuranosyl) purine (i6A) and its 2-methylthio derivative (ms2i6A), substantial amounts of cis- and trans-ribosylzeatin (io6A) and cis-2-(methylthio) ribosylzeatin (c-ms2io6A) were present.
The only species present in tRNATrp was i6A; it contained no zeatin derivatives.
