Metabolism of PCB and polybrominated biphenyl ether (PBDE) flame-retardants alter their toxicological properties and these metabolites are known to interfere with the binding of thyroid hormone (TH) to transthyretin (TTR) in rodents and humans.

Environmentally relevant concentrations of selected polychlorinated biphenyl (PCB) and polybrominated diphenyl ether (PBDE) flame retardant congeners and their hydroxylated (OH) and methoxylated (MeO) analogues that can perturb thyroid hormone-dependent processes were comparatively examined with respect to competitive binding with thyroxine (T (4)) and 3,5,3'-triiodothyronine (T (3)) thyroid hormones (THs) on recombinant human and gull albumin and transthyretin transport proteins.

This indicated that the combination of the more thyroid hormone-like brominated diphenyl ether backbone (relative to the chlorinated biphenyl backbone), and in combination of having an OH-group, results in a more effective competitive ligand on gull TTR relative to both T (3) and T (4).

Drug Metab Dispos. 2009 Oct;37(10):2095-102. Epub 2009 Jul 9.

4-Hydroxy-2,2',3,4',5,5',6-heptachlorobiphenyl (4-OH-CB187) was selected as a major hydroxylated polychlorinated biphenyl metabolite detected from serum of wildlife and humans and was examined for its effect on level of serum thyroid hormone in mice.
Binding levels of [(125) I] T (4) to serum proteins after administration of [(125) I] T (4) were significantly changed in 4-OH-CB187-pretreated mice: a decrease in the level of serum [(125) I] T (4)-transthyretin (TTR) complex and an increase in the binding level of [(125) I] T (4) to serum albumin and thyroxine binding protein in both strains of mice.
