3677417 | Solomon LR: Studies on the mechanism of -mediated inhibition of rat liver transaminases. Clin Chim Acta. 1987 Sep 30;168(2):207-17. Incubation of mitochondria-depleted rat liver homogenates with 5 mmol/l at 37 degrees C for 1 h inhibited both and alanine aminotransferases by 30%. Inhibition was prevented by decreasing temperature to 4 degrees C or by preincubating homogenates with but was unaffected by cyanamide and methylpyrazole which block oxidation and reduction respectively. |

On the other hand, attempts to potentiate the toxicity of acrolein by the aldehyde dehydrogenase inhibitor cyanamide enhanced only the release of alanine aminotransferase. |

Alcohol Clin Exp Res. 2000 Apr;24(4 Suppl):97S-99S. Serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels obtained at 0, 4, 8, and 12 weeks of administration of each aversive drug were compared between the two alcoholic groups. |

Inhibition of alcohol dehydrogenase by 4-methylpyrazole or of aldehyde dehydrogenase by cyanamide totally abolished hepatotoxicity despite of a severalfold increase in concentration in the perfusate. |

3663401 | Solomon LR: Evidence for the generation of transaminase inhibitor (s) during metabolism by rat liver homogenates: a potential mechanism for toxicity. Biochem Med Metab Biol. 1987 Aug;38(1):9-18. When homogenates were incubated at 37 degrees with 50 mM for 1 hr, alanine aminotransferase decreased by 20%, while aspartate aminotransferase was unchanged. Cyanamide, an aldehyde dehydrogenase inhibitor, had little effect on -mediated transaminase inhibition. |

2886311 | Rikans LE: The oxidation of acrolein by rat liver aldehyde dehydrogenases. Drug Metab Dispos. 1987 May-Jun;15(3):356-62. The metabolism of acrolein by low Km aldehyde dehydrogenase activities was markedly depressed in mitochondrial or cytosolic fractions from rats pretreated with cyanamide (2 mg/kg for 1 hr) or disulfiram (100 mg/kg for 24 hr). Hepatotoxicity was assessed on the basis of elevated serum alanine aminotransferase and sorbitol dehydrogenase activities and the loss of microsomal cytochrome P-450. |
