RESULTS: In the first step, the suppressive effect of diflubenzuron, soybean trypsin inhibitor (STI) and chitinase on RGR was confirmed.

The chitin fibrils of the peritrophic membrane were a target for chitin effectors, including 1) chitinase, which hydrolyzes chitin fibers inside the peritrophic membrane; 2) calcofluor, which binds to chitin and destroys the peritrophic membrane mesh structure; and 3) diflubenzuron, which inhibits chitin synthesis.

16347718 Merker RI, Smit J: Characterization of the Adhesive Holdfast of Marine and Freshwater Caulobacters. Appl Environ Microbiol. 1988 Aug;54(8):2078-2085.

None of the glycolytic enzymes had an effect on marine caulobacter holdfasts, but chitinase and lysozyme (both attack oligomers of GlcNac) disrupted the holdfasts of those freshwater caulobacters that bound WGA.
Despite some similarity to chitin, holdfasts did not bind Calcofluor and no measurable effects on holdfast production were detectable after cell growth in the presence of diflubenzuron or polyoxin D, inhibitors of chitin synthesis in other systems.

16346973 Smucker RA, Simon SL: Some Effects of Diflubenzuron on Growth and Sporogenesis in Streptomyces spp. Appl Environ Microbiol. 1986 Jan;51(1):25-31.

Exposure to diflubenzuron resulted in small increases in exported protein and in a ca. 20% increase in chitinase in both Streptomyces spp.

20229097 Stara J, Erban T, Hubert J: The effect of chitin metabolic effectors on the population increase of stored product mites. Exp Appl Acarol. 2010 Mar 13.

The study tested the effect of the chitin metabolic effectors, teflubenzuron, diflubenzuron, and calcofluor, and a combination of a chitinase and soybean trypsin inhibitor (STI) on the population growth of eight species of stored product mites under laboratory conditions.

Strains were isolated from water samples amended with 10 and 50 microg/ml of nine pesticides: organochlorinated insecticides (aldrin and lindane), organophosphorous insecticides (dimetoate, methyl-parathion and methidation), s-triazine herbicides (simazine and atrazine), fungicide (captan) and diflubenzuron (1-(-4-chlorophenyl)-3-(2,6-difluorobenzoyl urea), a chitinase inhibitor.

The incorporation was partially inhibited (50-70%) by both polyoxin-D (apparent Ki 0.04 microM) and diflubenzuron (apparent Ki 5-8 microM).
A major part of this labelled product has been characterized as chitin by its insolubility in alkali, resistance to degradation by proteases and its susceptibility to digestion by chitinase and HCl.

6450211 Horst MN: The biosynthesis of crustacean chitin by a microsomal enzyme from larval brine shrimp. J Biol Chem. 1981 Feb 10;256(3):1412-9.

The product was identified as chitin by its resistance to extraction with alkali and high concentrations of urea and the liberation of chito-oligosaccharides by treatment with purified chitinases.
The enzyme requires Mg2+ for activity and is inhibited by UDP and diflubenzuron, but not by Polyoxin D.
