19364082 Alberti C: Cytoskeleton structure and dynamic behaviour: quick excursus from basic molecular mechanisms to some implications in cancer chemotherapy. Eur Rev Med Pharmacol Sci. 2009 Jan-Feb;13(1):13-21.

Some anticytoskeleton agents are under evaluation for their applications in tumor chemotherapy; benomyl, griseofulvin, sulfonamides, that are used as antimycotic and antimicrobial drugs, appear to have a powerful antitumor potential by targeting microtubule assembly dinamics, together with exhibiting, in comparison with taxane and Vinca alkaloids, a more limited toxicity.
Molecular motors that drive molecule cargo translocation along the cytoskeletal highway, either through endocytic or secretory-exocytic mechanisms, include kinesin and cytoplasmic dynein, traveling on microtubule, and myosin family members, traveling along actin meshwork.

10525539 Cottingham FR, Gheber L, Miller DL, Hoyt MA: Novel roles for saccharomyces cerevisiae mitotic spindle motors. J Cell Biol. 1999 Oct 18;147(2):335-50.

The single cytoplasmic dynein and five of the six kinesin-related proteins encoded by Saccharomyces cerevisiae participate in mitotic spindle function.
The cytoplasmic and nuclear roles of both these motors could be partially substituted for by the microtubule-destabilizing agent benomyl, suggesting that these motors perform an essential microtubule-destabilizing function.

11085270 Steinberg-Neifach O, Eshel D: Simultaneous expression of both MAT loci in haploid cells suppresses mutations in yeast microtubule motor genes. Mol Gen Genet. 2000 Oct;264(3):300-5.

The kinesin-related Cin8p and cytoplasmic dynein are microtubule-associated motor proteins required for anaphase spindle elongation in the yeast Saccharomyces cerevisiae.
The presence of the two MAT loci also rendered haploid cells more resistant to the antimicrotubule drug benomyl.

We predicted benomyl sensitivity and nuclear migration defects using congruence; these phenotypes were uncorrelated with direct synthetic lethality.
We also predicted YLL049W as a new member of the dynein-dynactin pathway and provided new supporting experimental evidence.

Furthermore, increased localization of the light chain of the minus end directed motor protein dynein was detected on the microtubules in the benomyl-treated cells indicating that the suppression of microtubule dynamics may influence the binding of dynein on the microtubules and dynein-mediated cargo transport.

The slow growth of this dynein null mutant could be completely reverted to wild-type-like growth in the presence of benomyl, which can be explained by the observed redistribution of nuclei in the hyphal network.

In the presence of a low amount of benomyl, a microtubule-depolymerizing agent, however, a dynein mutant diploid strain exhibits a more pronounced chromosome loss phenotype than the control, indicating that cytoplasmic dynein plays a role in chromosome segregation.

Mutations in sldA and sldB also confer hypersensitivity to the microtubule-destabilizing drug benomyl. sldA and sldB were cloned by complementation of their mutant phenotypes using an A. nidulans autonomously replicating vector.

Disruption of MTs with benomyl and subsequent washout led to an equal distribution of the MTOC and random formation of highly motile and randomly oriented MTs throughout the cytoplasm.

9281582 Cottingham FR, Hoyt MA: Mitotic spindle positioning in Saccharomyces cerevisiae is accomplished by antagonistically acting microtubule motor proteins. J Cell Biol. 1997 Sep 8;138(5):1041-53.

The single dynein motor encoded by the S. cerevisiae genome performs an important but nonessential spindle-positioning role.
Microtubules in kip2Delta cells were shorter and more sensitive to benomyl than wild-type, in contrast to the longer and benomyl-resistant microtubules found in dyn1Delta and kip3Delta cells.

These dynamics of GFP-NUDA were not observed in cells treated with a microtubule-destabilizing drug, benomyl, suggesting they are microtubule-dependent.

Both the gradient and the Golgi apparatus were sensitive to brefeldin A or benomyl treatment, suggesting that the gradient represents microtubule-dependent vesicle trafficking between ER and Golgi.

Microtubules and cytoplasmic dynein, a microtubule-dependent motor, are required for nuclei to move along the hyphae of filamentous fungi.
The mutation alters microtubule staining and confers sensitivity to cold and benomyl, two treatments that destabilize microtubules.
