From P2 to P12 the frequencies of miniature EPSCs and miniature IPSCs in the Purkinje cells increased by 10-fold or more, suggesting progressive formation of functional synapses during this period.
Facilitatory effects of glycine were suppressed by strychnine (1 micro M), a specific blocker of the ionotropic glycine receptor, while the effects were also induced by other glycinergic agonists, including alpha-L-alanine (1 mM), L-serine (1 mM) and taurine (500 micro M).

We performed whole-cell recordings in 200-250 microm sections and immunocytochemical (ICC) studies in ventrobasal thalamus of rat brain (P12-P14).
Stimulation of medial lemniscus evoked inhibitory postsynaptic currents (IPSCs) which were purely glycinergic or GABA (A) ergic, or most commonly mixed glycinergic and GABA (A) ergic responses, based on abolition by strychnine, bicuculline, or combined antagonism.

GABAergic and glycinergic components were identified by blocking transmission with bicuculline and strychnine (SN), respectively.
In the medial limb of LSO, there was a dramatic change in the GABAergic IPSC component, decreasing from 78% at postnatal day 3 (P3)-P5 to 12% at P12-P16.

Administration of strychnine elicited synchronous activity bilaterally in lumbar ventral roots.
Rhythmic alternating locomotor-like activity could be produced by application of a combination of serotonin (5-HT), N-methyl-d-aspartate (NMDA), and dopamine in animals up to P12.

In rats (P12-P16) and in the presence of kynurenic acid, 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX) and bicuculline, ORG 24598 and ORG 25543 applied individually at a concentration of 10 microm induced a mean inward current of -10/-50 pA at -60 mV and increased significantly the decay time constants of miniature (mIPSCs), spontaneous (sIPSCs) and electrically evoked glycinergic (eIPSCs) inhibitory postsynaptic currents.
By contrast, the decay time constant, amplitude and frequency of miniature GABAergic IPSCs recorded in the presence of strychnine were not affected by ORG 24598 and ORG 25543.
