In the mammalian central nervous system, transporter-mediated reuptake may be critical for terminating the neurotransmitter action of D-serine at the strychnine insensitive glycine site of the NMDA receptor.
The Na (+) independent amino acid transporter alanine-serine-cysteine transporter 1 (Asc-1) has been proposed to account for synaptosomal d-serine uptake by virtue of its high affinity for D-serine and widespread neuronal expression throughout the brain.

In addition, excitatory amino acid transporters (EAAT) have a Cl (-) conductance which is gated by the joint binding of Na (+) and glutamate, but thermodynamically uncoupled to the flux of glutamate.
In these later cells, application of glutamate on the axon terminal evoked a current that reversed at E (Cl), was insensitive to bicuculline, TPMPA, strychnine, dl-AP5, CNQX and MCPG, but blocked by the glutamate transporter inhibitor dl-tBOA.

Picrotoxin, strychnine, and tetrodotoxin were used to isolate the effects of the glutamate analogs to the photoreceptor-bipolar cell synapse.
Under photopic conditions, the group III metabotropic glutamate receptor (mGluR) antagonist (RS)-alpha-cyclopropyl-4-phosphonophenylglycine (CPPG) only slightly reduced the b-wave, whereas the excitatory amino acid transporter (EAAT) blocker dl-threo-beta-benzyl-oxyaspartate (TBOA) removed most of it.

These sites are inherently distinct since the effects of strychnine and temperature on glycine binding are significantly different.
The relative activity of the uptake inhibitors indicate that in the adrenal chromaffin cell, glycine uptake is carried out by a System A amino acid transporter mechanism.

Specific agonists were puffed in the presence of cobalt, picrotoxin, and strychnine to identify glutamate receptors on these cells.
Most Cabs responded to both the alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionate (AMPA)/kainate receptor agonist kainate and the excitatory amino acid transporter (EAAT) substrate D-aspartate, and both responses were localized to the dendrites.

Glycine-evoked currents of brain macrophages were unaffected by strychnine, Cl (-)-free extracellular solution, N-[3-(4'-fluorophenyl)-3-(4'-phenylphenoxy) propyl]) sarcosine (NFPS) and amoxapine, but were abolished upon omission of extracellular Na (+).
