Conversely, mRNA of the proto-oncogene H-ras showed a decline of 35% after 3 days of rotenone treatment, and remained diminished for the duration of the experiment.

First, mitochondrial respiration, as measured by mitochondrial oxygen consumption, was suppressed in NIH-3T3 cells transformed with H-Ras (Q61L).
Second, oligomycin or rotenone did not reduce the cellular ATP levels in the H-Ras (Q61L) transformed cells, suggesting a diminished role of mitochondrial respiration in the cellular energy metabolism.

Superoxide generation was also inhibited by diphenylene iodonium, an inhibitor of flavoproteins, including NADPH oxidase, but not by rotenone, an inhibitor of the respiratory chain of the mitochondria.
